Product Description

A brand owner contacted us last year with a problem that should never have existed: their melatonin drops, labeled at 3mg per serving, tested at 1.1mg after just four months on the shelf. The contract manufacturer who produced them had never run a stability test on the finished product. The formula tasted fine. The active was gone.

This isn't unusual. A 2023 study published in JAMA found that 88% of melatonin products tested contained between 74% and 347% of their labeled dose. That study focused on gummies. In liquid drops, the problem is worse — because melatonin degrades faster in aqueous carriers, and every dropper squeeze adds another layer of dosing variance.

I've been working at our liquid supplement factory in Qufu since 2012. We've manufactured melatonin drops for brands across 50+ countries, and the single most common root cause of potency failure we see isn't raw material quality. It's pH.

Melatonin (N-acetyl-5-methoxytryptamine) is most stable in aqueous solution at pH 3.0–5.5. At pH above 7.4, the indole ring becomes electron-rich and undergoes oxidative cleavage, forming AFMK — a degradation product that is biologically inactive at melatonin receptors. Research published in the International Journal of Pharmaceutics showed that melatonin in pH 7 solution lost over 88% of its content within 14 days at room temperature. At pH 4, it was undetectable by day 21.

Here's the formulator's paradox: the acidic pH range that keeps melatonin stable produces a sour, harsh-tasting liquid. The neutral pH range that tastes pleasant destroys the active ingredient. Most contract manufacturers formulate for taste and never test what actually survives in the bottle.

This product page explains how we solve the stability-taste paradox in melatonin drops, how we verify dosing precision, and what customization options are available at Runxin.

The pH-Stability-Flavor Paradox in Melatonin Drops

This is the conversation every brand needs to have with their manufacturer — and most never do.

What Happens at the Wrong pH

At pH 6.0–7.4, melatonin in aqueous solution is "acceptable" for short-term stability (under 24 hours). This is the pH range most liquid supplement bases naturally fall into, because glycerin-water blends at neutral pH are the easiest to flavor and the most pleasant-tasting.

But a dietary supplement isn't consumed in 24 hours. A 30 mL bottle of melatonin drops at 1mg/mL, used at 1mL per night, sits on a consumer's nightstand for 30 days — often in a bathroom with humidity, temperature swings, and intermittent light exposure. Under those conditions, a pH 7 formulation loses potency continuously from the day it's manufactured.

At pH above 7.4, degradation accelerates dramatically. The indole nitrogen loses its proton, electron density increases, and the C2-C3 double bond undergoes oxidative cleavage. The product is AFMK (N1-acetyl-N2-formyl-5-methoxykynuramine), which does not bind to MT1/MT2 receptors. Your "melatonin" is chemically still present on an HPLC scan, but it's no longer functionally melatonin.

What Happens at the Right pH — and Why It's Hard

At pH 3.0–5.5, melatonin is maximally stable in aqueous solution. The protonated indole nitrogen resists oxidative attack, and degradation rates drop by an order of magnitude compared to neutral pH.

The problem: a pH 4 liquid tastes sour. Not mildly tart — genuinely unpleasant. Glycerin can mask some of this, and natural flavors help, but the underlying acidity is perceptible. Consumers who take melatonin drops before bed expect a soothing, pleasant experience. A sour, harsh liquid contradicts the product's entire positioning.

How Most Manufacturers Handle This

They formulate at pH 6–7, add flavors, and ship. If the brand asks about stability, they show raw material COA. They almost never test finished product potency at 3 months, 6 months, or 12 months. The brand doesn't ask because the brand doesn't know to ask.

How We Handle This

We formulate at pH 3.5–4.5 and build a flavor architecture that neutralizes the sour perception without raising the actual pH. This is not the same as adding a sweetener — it's a layered flavor system:

Layer

Function

Example

Acid-tamed base

Glycerin-dominant carrier with controlled water activity

65–75% glycerin matrix

Primary flavor

Covers the acid perception with a round, warm profile

Natural berry, chamomile-honey, vanilla

Secondary modulator

Binds to sour receptors without pH change

Specific natural flavor compounds

Sweetness balance

Provides perceived roundness without sugar spike

Stevia, monk fruit, or sucralose options

The result: a melatonin drop that tastes pleasant and maintains potency throughout its shelf life — because the pH was never compromised.

Dosing Precision: The Problem Nobody Measures

The 2023 JAMA study that found 88% of melatonin products were inaccurately labeled was a wake-up call for the industry. But it only measured gummies. In liquid drops, the dosing precision problem has two layers:

1. Degradation variance — As covered above, pH-related degradation means the actual melatonin content diverges from the label over time. A product that tests at 100% potency on day one may test at 55% after 90 days if formulated at the wrong pH.

2. Dropper variance — A standard dropper delivers a nominal volume per squeeze, but actual volume varies ±15–20% depending on viscosity, angle, squeeze force, and temperature. For a 1mg/mL product, this means each "1mg serving" could deliver 0.8–1.2mg on a good day — before accounting for any degradation.

Combined, a melatonin drop labeled at 3mg could realistically deliver anywhere from 1.5mg to 4.5mg depending on the age of the bottle and the consumer's dropper technique.

What we do differently:

  • We formulate at stability-optimized pH to minimize degradation variance
  • We specify calibrated droppers with documented drop size (e.g., 0.033mL per drop, 30 drops per mL)
  • We include dosing precision data in our formulation report, so brands can make accurate label claims
  • We test finished product potency at multiple time points, not just at release

Multi-Market Compliance: One Formulation, Different Rules

Melatonin's regulatory status varies more dramatically than almost any other supplement ingredient. This directly affects formulation and labeling decisions.

What this means for formulation: If you're building a global brand, you may need two or three formulation variants:

  • A US/Canada formula at your target dose (commonly 1–5mg)
  • An EU-specific formula at ≤2mg if you want to pursue food supplement registration in permissive member states
  • A separate regulatory strategy for UK, Australia, and Japan

We help brands plan formulation architecture for multi-market compliance before the first sample is produced. It's cheaper to design for multiple markets upfront than to reformulate after your first market entry.

Frequently Asked Questions

Q: Why does my melatonin drops label say 3mg but testing shows less?

A: The most common cause is pH-related degradation during storage. If the formulation was prepared at neutral pH for taste, melatonin degrades continuously in the liquid carrier. By the time the product reaches a consumer's hands — which can be months after manufacturing — actual potency may be significantly below the label claim. We formulate at pH 3.5–4.5 and verify finished product potency at multiple time points to prevent this.

Q: Can I use a clear bottle for my melatonin drops?

A: No. We will not produce melatonin drops in clear bottles for any order size. Melatonin is photosensitive — UV and visible light exposure causes photolytic degradation, forming AFMK which is inactive at melatonin receptors. Research shows potency losses of 20–30% within 30 days of light exposure. All our melatonin drop products use amber glass or UV-blocking PET.

Q: How do you control dosing accuracy in a liquid drop format?

A: Two layers. First, we formulate at stability-optimized pH so the melatonin content remains consistent throughout shelf life — this eliminates the biggest source of dosing variance. Second, we specify calibrated droppers with documented drop volume (e.g., 0.033mL per drop), and we include dosing precision data in every formulation report. Standard droppers have ±15–20% volume variance; our calibrated droppers reduce this to ±5–8%.

Q: My brand sells in the US and EU. Can I use one formulation for both markets?

A: Probably not. The US allows melatonin as a dietary supplement at any dose, while the EU classifies it as a prescription medicine in most member states. France permits food supplements at ≤2mg/day, but anything above requires pharmaceutical registration. We design formulation architectures for multi-market compliance before production begins, so you know which variants you need from the start.

Q: What is the MOQ for melatonin drops?

A: 50,000 bottles. This is based on our liquid production line's minimum efficient batch size for stability-verified formulation runs.

Q: What is the lead time?

A: Sample development: 10–15 working days (includes flavor optimization at target pH). Mass production: 20–25 working days after sample approval.
