Product Description

A brand owner came to us last year with a problem: their Vitamin E softgels turned from clear golden to dark amber within 4 months. Consumers were complaining about a "rancid smell." The formula was fine — 400 IU d-alpha-tocopherol, sunflower oil carrier. The issue was oxygen exposure during production.

At Runxin Biotech, we've manufactured Vitamin E capsules for 28 years. The lesson we learned early is this: Vitamin E is an antioxidant — it protects other compounds by sacrificing itself. If your production process exposes it to oxygen before it reaches the consumer, the product is already partially spent before it ships.

That's why every Vitamin E capsule we produce runs under nitrogen protection from mixing through encapsulation, and why our peroxide value control limits are tighter than GOED guidelines. The result: 36-month shelf life with consistent potency, not the 18-24 months you'll see from manufacturers who skip these steps.

Vitamin E Forms: Which One Fits Your Product?

Choosing the right form of Vitamin E affects stability, bioavailability, labeling, and cost. Here's what we recommend based on 28 years of production data:

Form

Source

Bioavailability

Stability

Best For

d-alpha-tocopherol

Natural (plant oil distillation)

Highest (100% relative activity)

Moderate — requires strict oxidation control

Premium natural positioning, US/EU retail

dl-alpha-tocopherol

Synthetic (petrochemical)

~74% of natural form

Moderate

Cost-effective mass market

Tocopheryl acetate

Esterified natural or synthetic

Lower (requires hydrolysis in gut)

Highest — ester bond protects phenolic OH

Long shelf life, warm-climate distribution

Mixed tocopherols (α/β/γ/δ)

Natural (soybean/sunflower distillation)

Broad-spectrum antioxidant

Good — gamma/delta are more oxidation-resistant

Full-spectrum positioning, clean label

Tocotrienols

Natural (palm/annatto/rice bran)

40-60x higher tissue penetration (research stage)

Moderate — heat-sensitive

Innovative/premium formulations

What this means for your brand: If your target market values "natural" and "high bioavailability," go with d-alpha-tocopherol and invest in proper oxidation control. If shelf life in hot climates is the priority, tocopheryl acetate gives you the longest runway. Mixed tocopherols with added gamma and delta fractions offer broader antioxidant coverage beyond what alpha alone provides.

The Oxidation Problem Nobody Talks About

Vitamin E has an ironic manufacturing challenge: it's an antioxidant, which means it's designed to react with oxygen. The problem is, once it reacts, it's gone.

Here's what happens during production without proper controls:

  1. During mixing: The Vitamin E oil is blended with carrier oil. Every minute of open-air mixing introduces dissolved oxygen.
  2. During transfer: Pumping the blend from mixing tank to encapsulation machine exposes it to headspace oxygen in pipes and hoppers.
  3. During encapsulation: The rotary die process generates heat (friction). Heat accelerates oxidation. Without nitrogen blanketing at the die, you're accelerating degradation at the most critical step.
  4. During drying: Softgels need 16-24 hours in drying tunnels. If the tunnel air isn't controlled, oxidation continues while the shell is still permeable.
  5. In the bottle: Residual headspace oxygen in the packaging continues the process, slowly, over months.

The result isn't immediately visible. The capsule looks fine. But the peroxide value has crept up. The p-anisidine value is rising. The tocopherol molecules that were supposed to protect the consumer's cells have already been consumed — by oxygen in your factory.

Our Oxidation Control Protocol

Here's how we handle Vitamin E production at Runxin, step by step:

Production Stage

Typical Manufacturer

Runxin Protocol

Raw material intake

Supplier COA review

COA review + in-house PV/p-AV testing before release

Mixing

Open vessel, ambient air

Closed vessel, nitrogen purged (O₂ < 1%)

Transfer to encapsulation

Open pipes, gravity feed

Sealed stainless lines, nitrogen blanket maintained

Encapsulation (rotary die)

Standard die, no atmosphere control

Nitrogen-flushed die enclosure, temperature monitored ≤ 25°C

Drying

Standard tunnel, ambient humidity

Controlled tunnel, ≤ 45% RH, temperature ≤ 25°C

Packaging

Standard filling, air headspace

Nitrogen-flushed headspace, amber or opaque bottles

Finished product testing

Assay + microbial

Assay + PV + p-AV + TOTOX + dissolution + microbial

Key oxidation limits we enforce:

Parameter

GOED Guideline

Runxin Internal Limit

Peroxide Value (PV)

≤ 5 meq/kg

≤ 3 meq/kg

p-Anisidine Value (p-AV)

≤ 20

≤ 10

TOTOX Value (2×PV + p-AV)

≤ 26

≤ 16

These aren't theoretical targets. We test every batch. Over the past 3 years, we've produced 150+ Vitamin E formulations. Only 2 batches required rework due to oxidation markers exceeding our internal limits — and both were caught before encapsulation.

Frequently Asked Questions

Q: Why does my Vitamin E product darken or smell rancid after a few months?

A: This is almost always oxidation. Vitamin E (tocopherol) is designed to react with oxygen — that's its function as an antioxidant. If your production process doesn't exclude oxygen during mixing, encapsulation, and packaging, the tocopherol molecules get consumed by factory oxygen before the product ever reaches your customer. The darkening is oxidized tocopherol turning into tocopheryl quinone. The smell is secondary oxidation products (aldehydes and ketones). The fix is nitrogen blanketing throughout production and inert gas flushing at packaging.

Q: What's the difference between d-alpha-tocopherol and dl-alpha-tocopherol?

A: d-alpha-tocopherol is the natural form, derived from plant oils. It has 100% relative biological activity. dl-alpha-tocopherol is the synthetic form, produced from petrochemical feedstock. It has approximately 74% of the biological activity of the natural form because it contains a mixture of 8 stereoisomers, of which only one matches the natural configuration. For premium or "natural" positioned products, d-alpha is the standard. For cost-sensitive mass market, dl-alpha is widely used. Both require the same oxidation controls during production.

Q: Can you make vegetarian Vitamin E capsules?

A: Yes. We offer HPMC (hydroxypropyl methylcellulose) vegetarian softgel shells as an alternative to gelatin. HPMC shells contain only 3-8% moisture (vs. 13-16% for gelatin), which actually gives a slight stability advantage for oxidation-sensitive fills like Vitamin E. The trade-off is that HPMC shells are more brittle and require adjusted drying parameters. We've been producing vegetarian Vitamin E softgels for clients targeting vegan, halal, and organic markets for over a decade.

Q: What's the typical lead time for a Vitamin E capsule order?

A: Sample production takes 5-7 working days. Mass production runs 15-25 working days depending on order size and packaging complexity. For first-time orders, add 3-5 days for formula confirmation and pilot batch validation. Our standard MOQ is 100,000 capsules, which is significantly lower than the industry norm of 300,000+.
